Predicting the Survival of Patients With Head and Neck Squamous Cell Carcinoma

An identified 9-gene prognostic signature based on the lysine (Kcr) regulators might contribute to prognosis stratification and treatment escalation for patients with head and neck squamous cell carcinoma (HNSCC).

Researchers aimed to establish and validate a Kcr-related prognostic signature of HNSCC and to assess the clinical predictive value of this signature by analyzing data from The Cancer Genome Atlas (TCGA). The mRNA expression profiles and clinicopathologic data were downloaded and the least absolute shrinkage and selection operator Cox regression model was used to generate the Kcr-related prognostic signature for HNSCC. The GSE65858 dataset from the Gene Expression Omnibus (GEO) database was used to validate the signature.

A 9-gene risk signature associated with the prognosis of HNSCC based on Kcr regulators was established to stratify HNSCC prognoses based on TCGA cohort. The risk signature effectively stratified patients into low- and high-risk groups. High-risk patients demonstrated poorer overall survival than the low-risk patients in the TCGA and GEO datasets. The 9-gene risk signature was more accurate for predicting the 5-year overall survival than other clinical parameters.

“We established a 9-gene prognostic signature based on the Kcr regulators and validated it in an external HNSCC cohort,” explained the study authors. “The constructed risk signature was an independent prognostic factor for HNSCC patients, which could effectively predict the survival of HNSCC patients and facilitate clinical decision-making for oncologists,” they added.
